Diffuse Axonal Injury

Diffuse axonal injury is a form of traumatic brain injury in which widespread damage to neuronal axons disrupts communication across the brain, often after rapid rotational or acceleration-deceleration forces. These forces stretch axonal membranes and cytoskeletal structures, impairing axonal transport and causing swelling, disconnection, and sometimes microscopic hemorrhages at sites such as the white matter, corpus callosum, and brainstem. Because the injury can be extensive despite limited findings on initial computed tomography, clinicians and researchers use magnetic resonance imaging, neuropathology, and neurological assessment to support diagnosis and prognosis. Studying diffuse axonal injury advances understanding of consciousness disorders, traumatic brain injury outcomes, and potential neuroprotective strategies.

Using Diffusion Tensor Imaging in Traumatic Brain Injury

Source: Laboratories of Jonas T. Kaplan and Sarah I. Gimbel—University of Southern California Traditional brain imaging techniques using MRI are very good at visualizing the gross structures of the brain. A structural brain image made with MRI provides high contrast of the borders between gray and white matter, and information about the size and shape of brain structures. However, these images do not detail the underlying structure and integrity of white matter networks in the brain, which...

Source: Li, Q., et al. Investigating Mammalian Axon Regeneration: In Vivo Electroporation of Adult Mouse Dorsal Root Ganglion. J. Vis. Exp. (2018)This video demonstrates the process of axonal regeneration in a mouse sciatic nerve following a crush injury. The isolated nerve is fixed and prepared for imaging, with fluorescent proteins expressed in neurons to visualize axon growth. Using an epifluorescence microscope, axon regeneration is traced from the injury site to assess the extent of...

This protocol presents the use of a dorsal root ganglion (DRG) injection with a viral vector and a concurrent dorsal root crush injury in an adult rat as a model to study sensory axon regeneration. This model is suitable for investigating the use of gene therapy to promote sensory axon regeneration.
