Horizontal Slice Preparation

Horizontal slice preparation is a neuroscience technique that produces thin sections of brain tissue cut parallel to the horizontal plane, preserving selected anatomical relationships for ex vivo study. After carefully isolating the brain region, researchers use a vibratome to section tissue in chilled, oxygenated artificial cerebrospinal fluid, which helps maintain cellular structure and physiological viability. The resulting slices support electrophysiological recording, fluorescence imaging, and circuit analysis, allowing investigators to examine synaptic transmission, neuronal connectivity, and network responses under controlled experimental conditions. This approach is especially valuable when the horizontal orientation retains pathways that may be disrupted in coronal or sagittal sections.

We developed a horizontal slice preparation of the adult mammalian retina with the plane of sectioning parallel to the retinal surface. Dendritic fields of nonradially oriented retinal neurons were not truncated, allowing the study of signal processing with patch-clamp and imaging techniques.
