Neuron Transduction

Neuron transduction is the process of introducing genetic material into neurons so they can produce a selected protein or exhibit a measurable molecular change. In neuroscience research, viral vectors such as adeno-associated viruses bind cell-surface receptors, enter neurons through endocytosis, and deliver an expression cassette that reaches the nucleus without necessarily replicating as a complete virus. Researchers use this approach to label neural circuits, manipulate neuronal activity, model disease-associated genes, and evaluate potential therapies. Transduction efficiency depends on vector design, serotype, delivery route, and neuronal type, making careful optimization essential for reproducible studies in cultured cells and living nervous systems.

Lentiviral-Mediated Transduction of Human Neurons for Tau Protein Aggregation Analysis

2025

This video demonstrates the process of transducing human neuronal cells with a lentivirus encoding a mutant human tau protein tagged with yellow fluorescent protein (YFP) to study protein aggregation. It outlines the steps from viral entry to mutant tau expression, culminating in the formation of visible cytoplasmic tau aggregates in the neurons.

Adult-born neurons of the olfactory bulb can be optogenetically controlled using Channelrhodopsin2-expressing lentiviral injection in the rostral migratory stream and chronic photostimulation with an implanted miniature LED.

Widespread Transduction of Mouse Neocortical Neurons by Subarachnoid Injection of AAV2

2025

A new technique for widespread delivery of adeno-associated virus that uses subarachnoid virus infusion is described. This method not only ensures widespread transduction of mouse neocortical neurons in superficial layers but also results in selective expression of the transgene in layer five pyramidal neurons, even when using a non-selective promoter.

Among the three main modes of HGT—transformation, conjugation, and transduction—transduction is unique in that it is mediated by bacteriophages, or bacterial viruses.Transduction occurs in two ways. Generalized transduction occurs during the lytic cycle of a bacteriophage infection. In this process, bacteriophages infect bacterial cells, replicate within them, and ultimately cause cell lysis, releasing newly assembled virions. Occasionally, random fragments of the bacterial genome are...

(2019).This video demonstrates viral transduction via in-utero injection in mouse embryos. A pregnant mouse is prepared for the surgery, and a midline incision is made to expose the uterine horns. A mixture of a virus containing the desired gene and a tracking dye is injected into the embryos within the uterine sacs at the...
