Optogenetic Tdp-43

Optogenetic TDP-43 is a light-controlled experimental approach for studying TAR DNA-binding protein 43 (TDP-43), a nuclear RNA-binding protein linked to neurodegenerative disease. In this method, TDP-43 is fused to a light-sensitive photoreceptor so that illumination, typically with blue light, promotes its rapid clustering or condensation in living cells, allowing researchers to control and observe changes in localization and assembly. This system helps investigate TDP-43 phase behavior, stress-granule interactions, RNA regulation, and toxicity in models of amyotrophic lateral sclerosis and frontotemporal dementia, while providing a precise framework for testing disease mechanisms and potential interventions.

Using atomic force microscopy in combination with biopanning technology we created a negative and positive biopanning system to acquire antibodies against disease-specific protein variants present in any biological material, even at low concentrations. We were successful in obtaining antibodies to TDP-43 protein variants involved in Amyotrophic Lateral Sclerosis.

This protocol describes the steps and data analysis required to successfully perform optogenetic functional magnetic resonance imaging (ofMRI). ofMRI is a novel technique that combines high-field fMRI readout with optogenetic stimulation, allowing for cell type-specific mapping of functional neural circuits and their dynamics across the whole living brain.
