Radiopaque Casting Agent

A radiopaque casting agent is a material introduced into anatomical spaces, especially blood vessels, to make their shape and connectivity visible with X-ray-based imaging. It works by filling a vascular network with a compound containing radiodense components, allowing the cast to attenuate X-rays and produce contrast while the material solidifies or remains stable within the specimen. In neuroscience, these agents help map cerebral arteries, veins, and microvascular pathways using angiography, computed tomography, or three-dimensional imaging. The resulting vascular models support studies of brain anatomy, development, cerebrovascular disease, and relationships between blood supply and neural function.

This protocol describes the preparation and characterization of a dendrimeric magnetic resonance imaging (MRI) contrast agent that carries cyclen-based macrocyclic chelates coordinating paramagnetic gadolinium ions. In a series of MRI experiments in vitro, this agent produced an amplified MRI signal when compared to the commercially available monomeric analogue.
