Reserve Pool

In neuroscience, the reserve pool is the population of synaptic vesicles held away from the active zone and recruited to sustain neurotransmitter release during prolonged or intense neuronal activity. When action potentials raise presynaptic calcium, vesicles in the readily releasable pool fuse with the plasma membrane; continued stimulation can mobilize reserve vesicles through activity-dependent trafficking, replenishing release sites and helping maintain synaptic transmission. Studying this pool clarifies how synapses balance rapid signaling with short-term plasticity, depression, and recovery. Its regulation is relevant to understanding information processing and disorders in which impaired vesicle cycling disrupts communication between neurons.

A live fluorescence imaging technique to quantify the replenishment and mobilisation of specific synaptic vesicle (SV) pools in central nerve terminals is described. Two rounds of SV recycling are monitored in the same nerve terminals providing an internal control.

In animals with large identified neurons (e.g. mollusks), analysis of motor pools is done using intracellular techniques1,2,3,4. Recently, we developed a technique to extracellularly stimulate and record individual neurons in Aplysia californica5. We now describe a protocol for using this technique to uniquely identify and characterize motor neurons within a motor pool.
