Serial Block-face Microscopy

Serial block-face microscopy is a three-dimensional imaging technique that reveals the ultrastructure of biological specimens by repeatedly imaging and removing successive tissue layers. In an electron microscope, a focused beam scans the exposed surface of an embedded sample, while an integrated ultramicrotome cuts away a thin section after each image; computational alignment then reconstructs the tissue volume. In neuroscience, this approach maps neurons, neurites, synapses, organelles, and other fine structures across large volumes of brain tissue. The resulting datasets support connectomics, circuit reconstruction, and quantitative studies of how cellular architecture relates to neural function and disease.
