Vesicle Recycling

Vesicle recycling is the cellular process that retrieves, renews, and reuses membrane-bound vesicles after they release their contents, helping cells sustain repeated secretion. In neuroscience, synaptic vesicles fuse with the presynaptic membrane during exocytosis, release neurotransmitters into the synaptic cleft, and are then recovered through endocytosis, often with clathrin-mediated membrane retrieval, before being refilled and returned to the readily releasable pool. This cycle preserves synaptic transmission during ongoing neuronal activity and regulates communication between neurons. Studying vesicle recycling helps explain synaptic plasticity, neural circuit function, and how disruptions in presynaptic trafficking may contribute to neurological disease.

We describe an optical assay for synaptic vesicle (SV) recycling in cultured neurons. Combining this protocol with double transfection to express a presynaptic marker and protein of interest allows us to locate presynaptic sites, their synaptic vesicle recycling capacity, and determine the role of the protein of interest.
