Minimum Alveolar Concentration

Minimum Alveolar Concentration (MAC) is the alveolar concentration of an inhaled anesthetic that prevents movement in response to a standardized surgical stimulus in 50% of patients, providing a comparative measure of anesthetic potency. At steady state, the anesthetic concentration in the alveoli reflects its partial pressure at the brain, where it alters neuronal signaling to suppress the perception of noxious stimuli and motor responses. Pharmacologists use MAC to compare volatile anesthetics, estimate effective dosing during general anesthesia, and interpret changes in anesthetic requirements across clinical conditions, supporting safer, more individualized control of unconsciousness and immobility.

The protocol presented herein seeks to evaluate the depth of anesthesia utilizing Dixon's Up-and-Down Design in sedated rats subjected to a standardized pain stimulus. Furthermore, it delineates a classification of the various movement patterns observed.

A Microtiter Plate Method to Assess the Minimum Inhibitory Concentration of an Antibiotic

This video demonstrates a microtiter plate-based method for determining the Minimum Inhibitory Concentration (MIC) of test antibiotics against bacteria. Upon adding a suspension of the bacterial pathogen Pseudomonas aeruginosa to a microtiter plate containing serial dilutions of a test antibiotic, and an inhibitor of the production of pyomelanin by the bacteria, the optical density of the wells is measured to compute the minimum inhibitory concentration of the antibiotic, and the impact of the...

Determining the Minimum Inhibitory Concentration of Enrofloxacin Against Avian Pathogenic E. coli

Begin with a suspension of avian pathogenic E. coli or APEC in growth media.APEC is a Gram-negative bacterium that infects poultry by entering through the respiratory tract. After breaching the mucosal layer, APEC adhere to epithelial cells, multiply locally, and disrupt the epithelial barrier to enter the bloodstream. The bacteria then spread to multiple organs, causing systemic infection.Add an equal volume of the bacterial suspension to each well of a multi-well plate containing increasing...

Gentamicin, an aminoglycoside antibiotic, is commonly administered via intermittent intravenous infusion to treat severe infections. An intermittent one-hour infusion of gentamicin, administered at eight-hour intervals, allows for precise control of plasma drug concentrations, minimizing toxicity while ensuring therapeutic efficacy. Pharmacokinetic principles govern the dynamics of plasma concentrations and can be mathematically described using specific equations. The plasma drug concentration...
