В этой статье описан метод изоляции ядер из свежезамороженной ткани человеческого мозга с использованием механических и центрифугальных методов. Процесс включает использование лизисного и сахарозного буферов для создания градиента плотности для эффективного разделения.
Isolating specific cell populations from complex tissues like the human brain is critical for de-risking target validation in neuroscience drug discovery. Fluorescence-activated nuclei sorting (FANS) enables high-purity isolation of nuclei from fresh-frozen tissue, supporting mechanistic studies of nuclear proteins and cellular heterogeneity. This approach improves predictive confidence in early discovery by providing reproducible, quantitative inputs for downstream assays and target engagement studies.
FANS integrates into the discovery continuum from target hypothesis testing through lead identification and preclinical validation by delivering purified nuclear fractions for functional and biochemical analysis.
