Microaspirazione viene eseguita tirando un capillare di vetro per creare una micropipetta con una punta molto piccola (2-50 micron di diametro a seconda delle dimensioni di una cellula o di un campione di tessuto), che viene poi collegato ad un trasduttore di pressione pneumatica e portato a termine prossimità di una cellula al microscopio. Quando l'punta della pipetta tocca una cella, una fase di pressione negativa è applicata alla pipetta dal trasduttore di pressione pneumatica di generazione di pressione ben definito sulla membrana cellulare.
