Acute hippocampal slices have been used for decades by neuroscientists to study neuronal circuits and synaptic properties. This technique has yielded major discoveries, such as long-term potentiation and other synaptic plasticity rules, and is still the most commonly used brain slice preparation. Due to its extensive and long-term use many different ways to prepare hippocampal slices exist today. While some laboratories favor coronal slices, others prefer to slice horizontally or to dissect the hippocampus in order to prepare transverse slices. Each of the slicing techniques presents its own specific set of advantages; such as retaining the entorhinal cortex connection, the possibility to obtain slices from the dorsal and ventral hippocampus, or to record sharp-wave ripples. However, the variety in slice preparation techniques also leads to variation in results, making the comparison between publications using different preparations difficult for non-expert readers. This collection will describe the major ways to obtain acute hippocampal slices. Laboratories using distinct hippocampal slice preparations are invited to participate. Being able to visualize how to prepare a specific type of hippocampal slices will help in reproducing other laboratories’ results and avoid costly travel. This collection will also offer guidance as to which technique is best suited for the experimenter’s needs, as well as advice on the inherent difficulties when comparing the results obtained from different preparations.
