Cholinergic Interneurons

Cholinergic interneurons are neurons that release acetylcholine and shape communication within neural circuits, making them important regulators of brain function and behavior. They influence nearby neurons through acetylcholine receptors, modifying synaptic transmission, neuronal excitability, and the balance between circuit activity and inhibition; in the striatum, their activity is coordinated with sensory and motor signals. By regulating circuits involved in movement, motivation, learning, and reward, cholinergic interneurons help determine how animals respond to environmental cues and select actions. Studying their signaling provides insight into normal behavior and disorders involving basal ganglia circuit dysfunction.

This video demonstrates the process of visualizing a biocytin-filled interneuron in a rat hippocampal section, highlighting its neurochemical identity through the use of specific antibodies and fluorescence microscopy.

Establishing a Co-Culture of Interneurons with Periventricular Endothelial Cells

2025

This video demonstrates a technique for establishing a co-culture of interneurons with perivascular endothelial cells. The cells are added to a one-well insert placed inside a culture dish. After the cells adhere to the dish, the insert is removed, allowing the cells to proliferate and interact and form a co-culture.

Assessing Interneuron Response to Chemoattractive Cues from Periventricular Endothelial Cells

2025

This video shows a method to test interneuron migration in response to chemoattractive cues from periventricular endothelial cells (PVECs). Interneurons are placed in a central compartment, with PVECs and control cells on either side. Increased migration towards PVECs indicates a positive response to their chemoattractive signals.

(2018)This video demonstrates the transplantation of interneuron precursor cells into the neonatal mouse brain using either a homotopic or heterotopic approach. The procedure involves securing an anesthetized recipient pup, identifying the target brain region, and injecting a suspension of precursor cells derived from a donor pup at the same developmental stage.

Labeling of Biocytin-Filled Interneurons in Rat Hippocampal Slices for Visualization
