Microinjection Diffusion Period

The microinjection diffusion period is the interval between delivering a substance into a localized tissue region and beginning behavioral measurements, allowing the injected solution to spread and act. During this time, the solute moves through the extracellular space, reaches nearby cellular targets, and undergoes concentration changes influenced by diffusion, uptake, and clearance. In behavioral neuroscience, selecting and consistently applying an appropriate diffusion period helps separate the intended pharmacological or neural effect from immediate effects of needle placement and fluid delivery. This timing is therefore important for reproducible experiments and for accurately linking localized brain manipulation to changes in behavior.

Zebrafish Microinjection Techniques

2023

One of the major advantages to working with zebrafish (Danio rerio) is that their genetics can be easily manipulated by microinjection of early stage embryos. Using this technique, solutions containing genetic material or knockdown constructs are delivered into the blastomeres: the embryonic cells sitting atop the yolk of the newly fertilized egg. Delivery into the cytoplasm is achieved either through direct injection into the blastomere, or via natural cytoplasmic movements that occur after a...

Diffusion in Agar ExpandNOTE: In this exercise, you will be given agar containing an indicator chemical called phenolphthalein. When phenolphthalein is exposed to the normal alkaline conditions in the agar, it will look pink. But when it is exposed to neutral or acidic conditions, it changes from pink to clear. You will make different size and shaped agar cubes as a model for cells to study the impact of cell size and shape on diffusion rate. To make the first set of cells, measure out and cut...

Cell Membranes and Diffusion In order to function, cells are required to move materials in and out of their cytoplasm via their cell membranes. These membranes are semipermeable, meaning that certain molecules are allowed to pass through, but not others. This movement of molecules is mediated by the phospholipid bilayer and its embedded proteins, some of which act as transport channels for molecules that otherwise would not be able to pass through the membrane, such as ions and carbohydrates.

Rodent Brain Microinjection Using a Quartz Microneedle

Source: Paolone, G., et al. Personalized Needles for Microinjections in the Rodent Brain. J. Vis. Exp. (2018).This video demonstrates a rodent brain microneedle injection using a quartz microneedle with holes. The microneedle is attached to a syringe and micropump to control the delivery rate. The needle with the injection solution is inserted into the brain with the delivery speed set at low. Once positioned, the delivery speed is increased. The needle is withdrawn after a brief pause.

Periodic Classification of the Elements

The periodic table arranges atoms based on increasing atomic number so that elements with the same chemical properties recur periodically. When their electron configurations are added to the table, a periodic recurrence of similar electron configurations in the outer shells of these elements is observed. Because they are in the outer shells of an atom, valence electrons play the most important role in chemical reactions. The outer electrons have the highest energy of the electrons in an atom...
