Script Concordance Test

The Script Concordance Test (SCT) is an assessment method that measures clinical reasoning when decisions must be made with incomplete, ambiguous, or evolving information. It presents short case vignettes in which a new finding changes the likelihood of a diagnostic hypothesis or the appropriateness of a management option, and examinees rate that change on a structured Likert scale. Responses are scored against the range of judgments provided by an expert reference panel, rewarding agreement with experienced reasoning rather than recall of a single answer. In health-professions education, SCTs assess decision-making under uncertainty and reveal how learners develop behaviorally relevant reasoning skills.

We present a protocol of personalized script-driven trauma-related imagery and clinical assessments within a comparison design for investigating peritraumatic dissociation (PD), psychophysiological reactions, i.e. heart rate (HR) and skin conductance (SC), and psychological features of often severely traumatized individuals with borderline personality disorder (BPD).

Detection of Rare Genomic Variants from Pooled Sequencing Using SPLINTER

Pooled DNA sequencing is a fast and cost-effective strategy to detect rare variants associated with complex phenotypes in large cohorts. Here we describe the computational analysis of pooled, next-generation sequencing of 32 cancer-related genes using the SPLINTER software package. This method is scalable, and applicable to any phenotype of interest.

The fenestrated liver sinusoidal endothelial cell is a biologically important filter system that is highly influenced by various diseases, toxins, and physiological states. These changes significantly impact on liver function. We describe methods for the standardisation of the measurement of the size and number of fenestrations in these cells.
