Startle Test

The startle test is a behavioral assay that measures an organism’s rapid, involuntary response to a sudden sensory stimulus, such as a loud sound. A brief stimulus activates sensory pathways and brainstem reflex circuits, producing measurable movements that can be recorded as response magnitude, latency, or habituation across repeated trials. Variations such as prepulse inhibition assess sensorimotor gating by testing whether a weaker warning stimulus reduces the reaction to a subsequent startling stimulus. In behavioral neuroscience, startle testing helps characterize attention, learning, stress reactivity, and neurological or psychiatric dysfunction in animal models and human research.

Habituation and Prepulse Inhibition of Acoustic Startle in Rodents

Habituation and prepulse inhibition of startle are operational measures of sensory gating. Sensory gating is disrupted in schizophrenia, and some other mental disorders and neurodegenerative diseases. We here describe a standard protocol to assess short-term and long-term habituation as well as prepulse inhibition of acoustic startle responses in rats and mice.

We describe a screening system's workflow and data analysis for evaluating chemical compound toxicity based on the zebrafish embryo vibration startle response. The system records the movements of zebrafish embryos upon exposure to a vibration stimulus and allows for an integrated evaluation of general toxicity/lethality and neuromuscular toxicity.

A fast and inexpensive method for the behavioral determination of hearing parameters like hearing thresholds, hearing impairments or phantom perceptions (subjective tinnitus) is described. It uses pre-pulse inhibition of the acoustic startle response and can be easily implemented in a personal computer using a programmable AD/DA-converter and a piezo sensor.
