Mlkl Fiber Characterization

Mlkl fiber characterization is the biochemical analysis of higher-order assemblies formed by mixed lineage kinase domain-like protein (MLKL), a key effector of necroptotic cell death. During necroptosis, upstream signaling activates MLKL through phosphorylation, promoting oligomerization and the formation of membrane-disrupting fibers or polymers. Characterizing these structures can assess their size, morphology, stability, and membrane-binding behavior using approaches such as protein purification, sedimentation, microscopy, and biochemical fractionation. These measurements help clarify how MLKL converts a regulated signaling event into plasma-membrane disruption and support investigation of necroptosis in inflammation, infection, tissue injury, and therapeutic development.

In Vivo Single-Fiber Recording of Sciatic Nerve C-Fibers in Rats

2025

This video demonstrates a single-fiber recording of C-fiber nerve conduction in rats. An anesthetized rat is incised to expose the sciatic nerve, which provides sensory and motor supply to the lower limbs. The individual C-fibers are then exposed. Electrical stimulation is provided using stimulus electrodes implanted in the foot, and a response is recorded from a single nerve fiber.
