Nrd1 Rbp

Nrd1 RNA-binding protein (Nrd1 Rbp) is a eukaryotic RNA-binding factor that helps recognize and regulate newly synthesized transcripts, particularly in transcription termination. Through its RNA recognition motif, Nrd1 binds specific sequence elements in nascent RNA and associates with Nab3 and the helicase Sen1 to form a termination complex; its interaction with RNA polymerase II also helps position this machinery on the transcription complex. This pathway directs termination of many non-polyadenylated transcripts and promotes their processing or degradation by the nuclear exosome. Studying Nrd1 Rbp provides insight into RNA surveillance, gene expression control, and the biochemical coordination of transcription and RNA processing.

Reporter Gene Repression Assay to Study Translational Regulation of a Target Gene

2025

This video demonstrates an in vivo assay in bacteria to study the interaction of RNA-binding proteins (RBPs) with RNA. The bacterial cells are transformed with two plasmid constructs — a binding-site plasmid encoding an mRNA containing a fluorescent reporter gene downstream of an RBP-binding site — while an RBP plasmid expresses the RBP proteins under the control of an inducer. Upon inducer-mediated increase in RBP production, the RBPs bind to the mRNA, resulting in translational repression of...

Here we are presenting a chromatin immunoprecipitation (ChIP) procedure for genome-wide location analysis of protein isoforms that differ in a histone-binding domain. We are applying it to ChIP-Seq analysis to identify the targets of the KDM5A/JARID1A/RBP2 histone demethylase.
