Protein-to-phospholipid Ratio

Protein-to-phospholipid ratio is a compositional measure that compares the amount of membrane-associated protein with the amount of phospholipid, helping characterize membrane organization and function. The ratio reflects how densely proteins are embedded in or attached to the lipid bilayer: changing protein abundance relative to phospholipids can alter membrane packing, fluidity, permeability, and the space available for transporters, receptors, and enzymes. In biochemistry, researchers use this measure to compare biological membranes, assess membrane purification or fractionation, and design or evaluate reconstituted lipid-protein systems. Interpreting the ratio alongside lipid composition and protein identity helps connect membrane composition with energy conversion, signaling, transport, and other cellular activities.

Quantitative Flow Cytometry to Study Labeled Protein-Phospholipid Vesicle Interactions

In this video, we demonstrate the interaction between proteins and artificial phospholipid vesicles using quantitative flow cytometry. The binding of fluorescently-labeled proteins to fluorescently-labeled phospholipid vesicles increases the mean fluorescence intensity, and this increase is detected by a flow cytometer.

Extraction and Analysis of Microbial Phospholipid Fatty Acids in Soils

Phospholipid fatty acids provide information about the structure of soil microbial communities. We present methods for extraction from soil samples with a single-phase chloroform mixture, fractionation of extracted lipids using solid phase extraction columns, and methanolysis to produce fatty acid methyl esters, which are analyzed by capillary gas chromatography.
