Assay Development Support

Assay development support encompasses the design, optimization, and validation of analytical tests used to measure biological or biochemical targets. In bioengineering, this process typically defines the analyte and detection principle, then tunes reagents, sample conditions, controls, and readout parameters to improve sensitivity, specificity, accuracy, and reproducibility. We describe a method to quantify the aggregation of misfolded proteins. Our protocol details lentiviral induced stable cell line generation, automated confocal imaging, and image analysis of protein aggregates. As an illustrative application, we studied the effect of small molecules in promoting SOD1 aggregation in a time- and dose-dependent manner.

In this video, we demonstrate protein-phosphoinositide interactions in a microfluidic pattern using the pH modulation assay, using pH-sensitive fluorescent dye tagged to phosphatidylethanolamine lipid of a supported lipid bilayer, called a microfluidic platform. The modulations in pH after protein binding to phospholipid cause fluorescence quenching.
