Cell Coverage Analysis

Cell Coverage Analysis is a quantitative imaging method that measures the proportion of a surface occupied by cells, providing an indicator of cell attachment, spreading, and growth. In bioengineering, microscopy images are processed by distinguishing cell-covered regions from the underlying substrate, often through image segmentation and thresholding, then calculating coverage as the cell-positive area relative to the total analyzed area. This measurement supports evaluation of biomaterials, tissue-engineered scaffolds, cell culture platforms, and engineered surfaces. Comparing coverage across conditions can reveal how substrate properties, culture treatments, or environmental factors influence cellular behavior and help guide the design of regenerative and diagnostic systems.

Human tumor xenografts are vascularized and infiltrated by cells of mouse origin during the growth phase in vivo. To circumvent the bias caused by these contaminating cells during downstream analysis, we developed a method allowing for comprehensive depletion of all mouse cells by magnetic cell sorting.

Heterokaryon Technique for Analysis of Cell Type-specific Localization

A flexible and efficient method for the characterization of cell type-specific protein localization and nucleocytoplasmic shuttling is described. This heterokaryon approach uses fluorescently-labeled fusion proteins to image protein localizations after cell fusion. The protocol is amenable to steady-state localizations or more dynamic determinations based on live cell imaging.
