Endothelial Medium Perfusion

Endothelial medium perfusion is a bioengineering method that continuously circulates culture medium over or through endothelial cell constructs to recreate key features of vascular environments. A pump or fluidic system controls the medium’s flow, supplying nutrients and oxygen while exposing cells to fluid shear stress, a mechanical cue that influences alignment, barrier function, and vascular signaling. This approach supports the development of in vitro blood vessel models, organ-on-chip systems, and engineered tissues with more physiologically relevant conditions than static culture. It also enables studies of vascular disease, drug transport, permeability, and endothelial responses to controlled changes in flow.

The goal of this protocol is to obtain high viability and high yield of hepatocytes and sinusoidal endothelial cells from liver. This is accomplished by perfusing the liver with a type IV collagenase solution via the portal vein, followed by differential centrifugation to obtain hepatocytes and sinusoidal endothelial cells.

This article describes a simple method to isolate and expand endothelial cells from the mouse aorta without using any special equipment. Our protocol provides an effective means of identifying mechanisms in endothelial cell physiopathology.

We provide a method for isolating and culturing pure populations of heart valve endothelial cells (VEC). VEC can be isolated from either side of the cusp or leaflet and immediately following, underlying interstitial cell (VIC) isolation is straightforward.
