Experimental Valve Procedures

Experimental valve procedures are emerging approaches for repairing, replacing, or evaluating heart valves through bioengineered devices, materials, and techniques. They work by restoring one-way blood flow: a native or engineered valve opens when pressure rises upstream and closes when pressure reverses, while researchers assess motion, durability, sealing, blood compatibility, and tissue response under controlled conditions. In bioengineering, these procedures support the development of prosthetic valves, minimally invasive delivery systems, and tissue-engineered replacements through bench testing, computational modeling, and preclinical studies. Their outcomes can improve treatment options for valve disease and guide safer, more durable cardiovascular implants.

This protocol describes the isolation, culture, and calcification of rat-derived valve interstitial cells, a highly physiological in vitro model of calcific aortic valve disease (CAVD). Exploitation of this rat model facilitates CAVD research in exploring the cell and molecular mechanisms that underlie this complex pathological process.
