Glucose Lactate Ph Sensing

Glucose, lactate, and pH sensing refers to the measurement of key chemical indicators that reflect metabolism, cellular activity, and physiological status. In bioengineering devices, glucose and lactate are commonly detected through enzyme-based reactions, such as glucose oxidase or lactate oxidase converting each analyte into electrochemically measurable products, while pH sensors respond to changes in hydrogen-ion concentration. Integrating these sensors enables simultaneous, real-time monitoring in biological fluids, cell cultures, tissue models, and wearable systems. The resulting data can help characterize metabolic changes, assess bioprocess performance, support disease monitoring, and guide the development of closed-loop biomedical technologies.

Membrane Potential Dye Imaging of Ventromedial Hypothalamus Neurons From Adult Mice to Study Glucose Sensing

The activity of single neurons from adult-aged mice can be studied by dissociating neurons from specific brain regions and using fluorescent membrane potential dye imaging. By testing responses to changes in glucose, this technique can be used to study the glucose sensitivity of adult ventromedial hypothalamic neurons.

The device is being developed for in situ sampling and electrochemical analysis of multiple analytes in a rapid and selective manner. This video describes a colorimetric assay for lactate measurement in cell lysates by using the lactate dehydrogenase enzyme. This enzyme converts lactate into pyruvate, along with the conversion of NAD to NADH that reacts with tetrazolium dyes and gives a colored product that can be measured spectrophotometrically to quantitate the lactate.
