Highly Inclined Swept Tile Microscopy

Highly Inclined Swept Tile Microscopy is an optical imaging technique that combines highly inclined illumination with tiled image acquisition to capture high-resolution views across large biological specimens. A thin excitation beam is swept through the sample at a steep angle, producing optical sectioning while limiting out-of-focus fluorescence; adjacent image tiles are then collected and assembled into a larger field of view. In bioengineering, this approach supports visualization of cells, tissues, biomaterials, and engineered constructs with improved spatial coverage and reduced imaging burden. It can help relate cellular organization to tissue-scale structure in studies of development, disease, and regenerative engineering.

High-resolution intravital imaging with enhanced contrast up to 120 µm depth in lymph nodes of adult mice is achieved by spatially modulating the excitation pattern of a multi-focal two-photon microscope. In 100 µm depth we measured resolutions of 487 nm (lateral) and 551 nm (axial), thus circumventing scattering and diffraction limits.
