Ht 29 Cells

HT-29 cells are a human colorectal adenocarcinoma cell line used as an in vitro model of intestinal epithelial biology and cancer, making them valuable in bioengineering research. When cultured under appropriate conditions, these adherent epithelial cells can proliferate and, in selected sublines or differentiation protocols, develop features such as mucin production and more specialized intestinal phenotypes. Researchers use HT-29 cells to build engineered gut models, evaluate drug transport and toxicity, study tumor-associated epithelial behavior, and test biomaterials or tissue interfaces. Their reproducible growth and human origin support controlled experiments, although results require careful interpretation because the cells retain cancer-associated characteristics.

This paper presents a sensitive method called Circle-Seq for purifying extrachromosomal circular DNA (eccDNA). The method encompasses column purification, removal of remaining linear chromosomal DNA, rolling-circle amplification and high-throughput sequencing. Circle-Seq is applicable to genome-scale screening of eukaryotic eccDNA and studying genome instability and copy-number variation.

Source: Poore, K., et al. Epithelial Cell Infection Analyses with Shigella. J. Vis. Exp. (2024)This video demonstrates an in vitro assay to assess Shigella invasion and intracellular replication in human colonic epithelial monolayers. It highlights steps for infection, antibiotic selection, host cell lysis, and recovery of intracellular bacteria for downstream analysis.

2026

Source: Rustad, M., et al. Synthesis of Infectious Bacteriophages in an E. coli-based Cell-free Expression System. J. Vis. Exp. (2017).This video demonstrates the synthesis of bacteriophages using a cell-free transcription–translation (TXTL) system. A master mix is prepared using a bacterial crude extract, an energy mix, an amino acid mix, an inhibitor for a DNA-degrading enzyme, and bacteriophage DNA, then incubated. The in vitro molecular machinery enables viral protein expression and DNA...
