Human Adipose Tissue Stromal Cells

Human adipose tissue stromal cells are heterogeneous, multipotent cells isolated from the stromal vascular fraction of adipose tissue and studied for their roles in repair and tissue engineering. After enzymatic digestion and removal of mature adipocytes, the resulting cells attach to culture surfaces and respond to biochemical and mechanical cues through paracrine signaling, extracellular-matrix remodeling, and lineage-specific differentiation. In bioengineering, researchers expand these cells, characterize their surface markers and potency, and combine them with biomaterials or three-dimensional scaffolds to support adipose, bone, cartilage, and vascular tissue models. Their accessibility and regenerative potential make them valuable for developing engineered tissues and cell-based therapies.

Adipose tissue (AT) is a site of intense immune cell activation and interaction. Almost all cells of the immune system are present in AT and their ratios are altered by obesity. Proper isolation, quantification, and characterization of AT immune cell populations are critical for understanding their role in immunometabolic disease.

This protocol describes the isolation of pig adipose-derived stem cells (pADSC) from subcutaneous adipose tissues with examination of multipotency. The multipotent pADSC are used to delineate processes of adipocyte differentiation and study transdifferentiation into multiple cell lineages of mesodermal mesenchyme or further lineages of ectoderm and endoderm for regenerative studies.
