Hydrogel Sheet Manipulation

Hydrogel sheet manipulation is the controlled handling, positioning, and shaping of thin, water-rich polymer networks for bioengineering applications. Because hydrogels combine a hydrated three-dimensional matrix with limited mechanical strength, manipulation requires careful control of adhesion, hydration, deformation, and applied force to prevent tearing, wrinkling, or loss of structure. Researchers use these procedures to assemble layered constructs, transfer cell-laden materials, and prepare hydrogel sheets for tissue models, drug delivery systems, and regenerative engineering. Reliable manipulation improves geometric accuracy and preserves the biological and material properties needed for reproducible fabrication, characterization, and integration with surrounding tissues or devices.

Construction of Modular Hydrogel Sheets for Micropatterned Macro-scaled 3D Cellular Architecture

We describe the fabrication of micropatterned hydrogel sheets using a simple process, which can be assembled and manipulated in a freestanding form. Using these modular hydrogel sheets, a simple macro-scaled 3D cell culture system can be generated with a controlled cellular microenvironment.

Easy Manipulation of Architectures in Protein-based Hydrogels for Cell Culture Applications

Different methods to manipulate three-dimensional architecture in protein-based hydrogels are evaluated here with respect to material properties. The macroporous networks are functionalized with a cell-adhesive peptide, and their feasibility in cell culture is evaluated using two different model cell lines.

For creation of highly organized structures of complex tissue, one must assemble multiple material and cell types into an integrated composite. This combinatorial design incorporates organ-specific layered cell sheets with two distinct biologically-derived materials containing a strong fibrous matrix base, and endothelial cells for enhancing new vessels formation.
