Microfluidic Chip Fabrication

Microfluidic chip fabrication is the process of creating miniature devices with precisely engineered channels that manipulate microliter- to nanoliter-scale fluids, enabling controlled analysis with small samples and reagent volumes. In bioengineering, fabrication commonly uses photolithography to pattern a mold, followed by soft lithography to replicate the channel design in polymers such as polydimethylsiloxane (PDMS); plasma bonding then seals the patterned layer to glass or another substrate. These chips support cell culture, biochemical assays, tissue models, and point-of-care diagnostics by integrating fluid handling, mixing, separation, and detection in compact platforms. Advances in materials and manufacturing continue to improve device robustness, scalability, and compatibility with biological systems.

We present a discrete droplet sample introduction system for inductively coupled plasma mass spectrometry (ICPMS). It is based on a cheap and disposable microfluidic chip that generates highly monodisperse droplets in a size range of 40−60 µm at frequencies from 90 to 7,000 Hz.

Here we demonstrate how to fabricate thermoplastic microfluidic chips using hot embossing and heat sealing. Then we demonstrate how to use in situ light directed surface grafting and polymerization through the sealed chip to form the composite solid phase columns.

A microfluidic biosensor platform was designed and fabricated using low-cost dry film photoresist technology for the rapid and sensitive quantification of various analytes. This single-use system allows for the electrochemical readout of on-chip-immobilized enzyme-linked assays by means of the stop-flow technique.
