Micromanipulator Alignment

Micromanipulator alignment is the process of positioning and calibrating a precision instrument so its movement and working tip correspond accurately with the microscope’s optical field and the sample’s coordinate system. In bioengineering, alignment typically involves centering the tool, adjusting its height and angle, and verifying that movement along each control axis produces predictable tip displacement without unwanted drift. Proper alignment improves positional accuracy, repeatability, and control during microscale operations such as cell injection, embryo manipulation, microassembly, and tissue handling. It also reduces sample damage and measurement error, supporting reliable experiments in cell engineering, reproductive technologies, microfluidics, and biomedical device development.

Microiontophoresis entails movement of ions from a micropipette in response to a difference in electrical potential between the inside and outside of the micropipette. Biologically active molecules are thereby delivered in proportion to electrical current. We illustrate acetylcholine microiontophoresis in conjunction with micromanipulation to study endothelium-dependent vasodilation in the microcirculation.
