Pancreas Decellularization

Pancreas decellularization is a bioengineering technique that removes cells from pancreatic tissue while preserving its extracellular matrix, three-dimensional architecture, and biochemical cues. The process typically uses perfusion or immersion with detergents, enzymes, and rinsing steps to lyse cells and clear DNA and other cellular components, while limiting damage to the matrix and vascular structure. The resulting acellular scaffold can support the study of pancreatic microenvironments and guide the development of engineered tissues for diabetes research. It also provides a platform for recellularization with pancreatic or stem-cell-derived cells, advancing regenerative medicine, disease modeling, and potential islet transplantation strategies.

Isolation and Decellularization of a Whole Porcine Pancreas

Tissue engineering of the whole pancreas is a challenge because of its exocrine and endocrine functions. We show a method for the dissection of an intact porcine pancreas and the process of successful decellularization by perfusion of detergents Triton X-100, sodium deoxycholate, and deoxyribonuclease.

Decellularization and Recellularization of Whole Livers

Perfusion decellularization is a novel technique to produce whole liver scaffolds that retains the organ's extracellular matrix composition and microarchitecture. Herein, the method of preparing whole organ scaffolds using perfusion decellularization and subsequent repopulation with hepatocytes is described. Functional and transplantable liver grafts can be generated using this technique.
