Peptoid Synthesis

Peptoid synthesis is the chemical production of peptoids, peptide-inspired polymers in which side chains attach to the amide nitrogen rather than the conventional alpha carbon. A widely used submonomer method builds sequences stepwise by acylating a growing chain with bromoacetic acid, then displacing the bromide with a primary amine; repeating these cycles provides control over composition and length. After cleavage and purification, the resulting peptoids can be tailored for molecular recognition, antimicrobial activity, biomaterial design, and drug delivery. Their resistance to enzymatic degradation and broad chemical versatility make them valuable building blocks for bioengineering and synthetic biology research.

A protocol to synthesize peptoids with mixed cationic functionality in the same sequence is presented (lysine- and arginine-type monomers). Subsequent testing of these compounds against Leishmania mexicana, the protozoan parasites that cause cutaneous leishmaniasis, is also described.

A simple and general manual peptoid synthesis method involving basic equipment and commercially available reagents is outlined, enabling peptoids to be easily synthesized in most laboratories. The synthesis, purification and characterization of an amphiphilic peptoid 36mer is described, as well as its self-assembly into highly-ordered nanosheets.
