Photo-convertible Proteins

Photo-convertible proteins are fluorescent or light-responsive proteins whose optical properties change after exposure to specific wavelengths, enabling controlled labeling and visualization in living systems. Light triggers photochemical changes in the protein’s chromophore, producing a reversible or irreversible shift in fluorescence color, intensity, or emission state. In bioengineering, these proteins support super-resolution microscopy, pulse-chase experiments, cell-lineage tracking, and studies of protein movement within cells. By allowing researchers to mark selected molecules or populations at defined times, photo-convertible proteins provide precise spatial and temporal information while reducing reliance on fixed samples and expanding the capabilities of live-cell imaging.

We established the photoconvertible PSmOrange system as a powerful, straight-forward and cost inexpensive tool for in vivo cell tracking in GFP transgenic backgrounds. This protocol describes its application in the zebrafish model system.

Here, we present a method for the photoactivated switch of photoconvertible fluorescent proteins (PCFPs) in the living zebrafish embryo and further tracking of photoconverted protein at specific time points during development. This methodology allows monitoring of cell biological events underlying different developmental processes in a live vertebrate organism.

The present protocol describes a procedure to perform fluorescent size exclusion chromatography (FSEC) on membrane proteins to assess their quality for downstream functional and structural analysis. Representative FSEC results collected for several G-protein coupled receptors (GPCRs) under detergent-solubilized and detergent-free conditions are presented.
