Polymer Microarray Technique

Polymer microarray technique is a high-throughput method for organizing and testing many distinct polymer compositions in spatially addressable microscopic spots on a single substrate, enabling rapid comparison of material properties and biological responses. In bioengineering, robotic deposition or in situ polymer formation creates an array in which each location contains a defined formulation; cells or biomolecules can then be exposed to the spots and evaluated for attachment, growth, morphology, or function. This approach accelerates biomaterial discovery by linking polymer chemistry with cell-material interactions, supporting the selection of coatings, scaffolds, and other engineered materials for tissue engineering, regenerative medicine, and related applications.

A technique called Comprehensive Microarray Polymer Profiling (CoMPP) for the characterisation of plant cell wall glycans is described. This method combines the specificity of monoclonal antibodies directed to defined glycan-epitopes with a miniature microarray analytical platform allowing screening of glycan occurrence in a broad range of biological contexts.

Source: Venkateswaran, S., et al. High-throughput Identification of Bacteria Repellent Polymers for Medical Devices. J. Vis. Exp. (2016)This video demonstrates a high-throughput method to identify bacteria-repellent polymers using a microarray slide exposed to mixed bacterial cultures. Bacterial binding is assessed by DAPI fluorescence, where reduced signal indicates polymers with minimal surface attachment.

This video demonstrates a method to detect the Cyanobacterial strains in an environmental sample using an antibody microarray chip. Samples carrying various bacteria interact with printed antibodies, and immunostaining with fluorophore-labeled anti-cyanobacterial-strain antibodies reveals the presence of specific cyanobacterial strains through fluorescence in distinct spots.

We describe a procedure for profiling salivary proteins using multiplexed microsphere-based antibody arrays. Monoclonal antibodies were covalently linked to fluorescent dye-encoded 4.5 μm polymer microspheres using carbodiimide chemistry. The modified microspheres were deposited in fiber-optic microwells to measure protein levels in saliva using fluorescence sandwich immunoassays.
