Refractive Index Detection

Refractive index detection is an optical measurement that identifies changes in how light travels through or interacts with a material, providing a label-free way to characterize composition and concentration. It works because dissolved molecules, particles, or binding events alter a medium’s refractive index; an instrument monitors the resulting change in beam refraction, phase, intensity, or resonance at an optical interface. In bioengineering, this principle supports biosensors, chromatography, and microfluidic assays for monitoring proteins, cells, metabolites, and molecular interactions. Its noninvasive, real-time readout can improve process monitoring and enable sensitive analysis when fluorescent or radioactive labels are undesirable.
