Reverse Molding

Reverse molding is a fabrication technique that reproduces the shape and surface features of a physical template by first creating a negative mold. The process typically involves casting a mold material around a master structure, removing the master after the material sets, and filling the resulting cavity with a biomaterial that cures or solidifies into a replica. In bioengineering, this approach enables precise production of complex scaffolds, microfluidic structures, and tissue-mimetic models that are difficult to manufacture directly. By preserving microscale geometry and reducing fabrication complexity, reverse molding supports studies of cell behavior, biomaterial performance, and regenerative design.

Printing Thermoresponsive Reverse Molds for the Creation of Patterned Two-component Hydrogels for 3D Cell Culture

A bioprinter was used to create patterned hydrogels based on a sacrificial mold. The poloxamer mold was backfilled with a second hydrogel and then eluted, leaving voids which were filled with a third hydrogel. This method uses fast elution and good printability of poloxamer to generate complex architectures from biopolymers.

We have devised a method for low-cost and rapid prototyping of liquid elastomer rubber injection molded devices by using fused deposition modeling 3D printers for mold design and a modified desiccator as a liquid injection system.

This article introduces a unique method for fabricating master molds with an air-vent hole to prevent air entrapment during viscous polymer casting, enabling the fabrication of gelatin microneedles with sharp tips. By reducing the casting volume of high-viscosity gelatin, thin-walled hollow pyramidal microneedles could also be fabricated.
