Reverse Phase Hplc

Reverse-phase high-performance liquid chromatography (RP-HPLC) is a separation technique that resolves compounds according to their hydrophobicity, making it valuable for analyzing and purifying biomolecules and bioengineering products. The method uses a nonpolar stationary phase, typically bonded silica, with a relatively polar mobile phase; compounds interact with the stationary phase to different degrees and are eluted as the solvent becomes more organic, often through a controlled gradient. RP-HPLC supports peptide and protein characterization, purity assessment, and purification of small-molecule therapeutics, providing retention-time and peak-area data for quality control, process development, and research.

Here, we describe in great detail an established and robust protocol for the extraction of glucosinolates from ground plant materials. After an on-column sulfatase treatment of the extracts, the desulfoglucosinolates are eluted and analyzed by high-pressure liquid chromatography.
