Spheroid Formation Methods

Spheroid formation methods are techniques used to assemble living cells into three-dimensional, tissue-like clusters that better reproduce aspects of native cell organization than flat monolayer cultures. They work by limiting cell attachment to a solid surface or guiding cells into close contact, allowing cell-cell adhesion, compaction, and extracellular matrix production to drive aggregate formation; common approaches include hanging drops, low-adhesion plates, rotating cultures, and microwells. In bioengineering, spheroids provide controllable models for studying tissue development, cell interactions, drug responses, and tumor biology. Their size, uniformity, and internal gradients can also support scaffold-free tissue fabrication and improve the design of organoids and regenerative therapies.

The uneven loss of medium from microtiter plates affects the reproducibility of uniform multicellular tumor spheroid formation. Improving culture conditions to reduce significant medium loss will improve the reproducibility of spheroid formation and the results of spheroid-based assays using the liquid-overlay technique.

Here, we describe a rapid and flexible protocol for the formation of 3D cell spheroids through cell aggregation. This is easily adapted to multiple cell types and is suitable for use in a variety of applications including cell migration, invasion, or anoikis assays, and for imaging and quantifying cell-matrix interactions.
