Wild-type P53 Delivery

Wild-type p53 delivery is a bioengineering strategy that introduces a functional TP53 gene or p53 protein into cells with absent, mutated, or inactive tumor-suppressor activity. Delivery systems such as viral vectors, nanoparticles, and engineered carriers transport p53 cargo into target cells, where restored p53 signaling can trigger cell-cycle arrest, DNA-damage responses, or apoptosis. This approach supports research on cancer biology, gene therapy, and precision drug delivery by testing how replacement of defective p53 influences tumor-cell behavior. Continued advances in carrier design, targeting, and controlled release may improve delivery efficiency while limiting effects on healthy tissues.

Wild-Type Blocking PCR to Detect Low-Frequency Somatic Mutations

2025

This video describes wild-type blocking polymerase chain reaction, or WTB-PCR, which detects somatic mutations. The PCR-based technique uses a locked nucleic acid, or LNA, an oligonucleotide that binds to its complementary wild-type allele and blocks its elongation. The blocking selectively allows the amplification of low-level mutant alleles over their wild-type variants in the sample.

(2018)This video demonstrates the cultivation and phenotypic comparison of wild-type and mutant Streptomyces coelicolor strains. It outlines procedures for minimizing spontaneous mutations, preventing cross-contamination, and promoting bacterial growth through proper plating techniques. Finally, the video highlights visual differences in colony morphology to illustrate the...

A PCR-based Genotyping Method to Distinguish Between Wild-type and Ornamental Varieties of Imperata cylindrica

2012

We provide a cost-effective and rapid molecular genotyping protocol that employs variety-specific PCR primers that target DNA sequence differences within the chloroplast trnL-F spacer region to differentiate between varieties of Imperata cylindrica (cogongrass) that cannot be distinguished by morphology alone. These varieties include the federally listed noxious weed, cogongrass and closely-related, wide-spread ornamental variety, I. cylindrica var. koenigii (Japanese blood grass).
