Femur Scan Selection

Femur scan selection is the process of choosing an imaging method that best answers a specific question about femoral structure, composition, or disease. The choice depends on how each technique generates contrast: radiography uses X-rays to show overall bone shape, computed tomography reconstructs cross-sectional anatomy, magnetic resonance imaging detects tissue-specific signals, and dual-energy X-ray absorptiometry estimates bone mineral density. In biological research and clinical assessment, selecting the appropriate scan improves visualization of fractures, cortical and trabecular bone, marrow, or mineral loss while balancing spatial detail, acquisition time, cost, and radiation exposure. Standardized selection also supports reliable comparisons across studies and longitudinal measurements.

The protocol describes a novel murine femur window chamber model that can be used to track movement of cells in the femoral bone marrow in vivo. Intravital multiphoton fluorescence microscopy is used to image three components of the femoral bone marrow (vasculature, collagen matrix, and neutrophils) over time.

One constraint of preclinical research in the field of bone repair is the lack of experimental control over the local mechanical environment within a healing bone lesion. We report the design and use of an external fixator for bone repair with the ability to change fixator stiffness in vivo.
