3d Epithelial Cell Models

3D epithelial cell models are laboratory systems that recreate the three-dimensional organization of epithelial tissues, which form protective and selective barriers throughout the body. Unlike conventional two-dimensional cultures, these models allow epithelial cells to interact with neighboring cells and an extracellular matrix while establishing spatial architecture, polarity, and tissue-like barriers. Researchers use them to study development, tissue function, barrier integrity, infection, disease mechanisms, and responses to therapeutic compounds. By more closely reflecting the structure and behavior of epithelial tissues in vivo, 3D models can provide biologically relevant insights and support advances in regenerative biology, drug evaluation, and personalized research.

Culturing and Applications of Rotating Wall Vessel Bioreactor Derived 3D Epithelial Cell Models

A rotating cell culture system that allows epithelial cells to grow under physiological conditions resulting in 3-D cellular aggregate formation is described. The aggregates generated display in vivo-like characteristics not observed in conventional culture models and serve as a more accurate organotypic model system for a multitude of scientific investigations.

Studying medullary thymic epithelial cells in vitro has been largely unsuccessful, as current 2D culture systems do not mimic the in vivo scenario. The 3D culture system described herein - a modified skin organotypic culture model - has proven superior in recapitulating mTEC proliferation, differentiation and maintenance of promiscuous gene expression.

Methods to Study Epithelial Transport Protein Function and Expression in Native Intestine and Caco-2 Cells Grown in 3D

We describe simple methods to study the regulation of intestinal serotonin transporter (SERT) function and expression using an in vitro cell culture model of Caco-2 cells grown in 3D and an ex vivo model of mouse intestines. These methods are applicable to the study of other epithelial transporters.

Here, we describe an in vitro protocol to model hypoxia/reoxygenation damage in the highly metabolic proximal tubular epithelial cells. This model is designed to induce metabolic stress-induced damage, which can be measured through the expression of proximal tubule damage markers and assessment of mitochondrial respiratory function.
