Absolute Quantification

Absolute quantification is the determination of the exact amount or concentration of a biological target in a sample, rather than its level relative to another sample or reference gene. It typically relies on standards with known quantities to generate a calibration curve, allowing a measured signal, such as fluorescence or ion intensity, to be converted into target copies, mass, or concentration while accounting for dilution and sample volume. In biology, absolute quantification supports measurements of nucleic acids, proteins, metabolites, cells, and microorganisms. It enables comparisons across experiments, validates biological models, and provides quantitative data for diagnostics, environmental monitoring, and therapeutic research.

This protocol describes how to perform absolute quantification assays of target proteins within complex biological samples using selected reaction monitoring. It was used to accurately quantify proteins of the mouse macrophage chemotaxis signaling pathway. Tandem splicing events occur at sites less than 12 nucleotides apart. Quantifying ratios of such splice variants is feasible using an absolute quantitative PCR approach.
