Acetylation Stoichiometry

Acetylation stoichiometry is the quantitative measurement of how many acetyl groups are attached to a protein or other biomolecule, revealing the extent of its modification. Acetyltransferases add acetyl groups, often to lysine residues, while deacetylases remove them; the measured ratio of modified to unmodified molecules can therefore reflect the balance of these opposing activities. In biology, stoichiometry helps distinguish abundant, functionally important acetylation from low-frequency events and clarifies how modification changes protein stability, localization, interactions, or activity. Applied to histones and other cellular proteins, it supports studies of chromatin regulation, gene expression, signaling, and disease-associated molecular changes.

Here, we present unbiased quantification of site-specific protein acetylation and/or succinylation occupancy (stoichiometry) of an entire proteome through a ratiometric analysis of endogenous modifications to modifications introduced after quantitative chemical acylation using stable isotope-labeled anhydrides. In combination with sensitive data-independent acquisition mass spectrometry, accurate site occupancy measurements are obtained.
