Umbilical Artery Flow

Umbilical artery flow is the movement of fetal blood through the umbilical arteries toward the placenta, where it supports exchange with the maternal circulation. Driven by fetal cardiac contraction, this flow carries relatively deoxygenated blood and metabolic waste to placental capillaries, while placental vascular resistance influences its speed and pattern. Doppler ultrasound measures umbilical artery blood-flow velocity and resistance, providing a noninvasive assessment of placental function and fetal well-being. In biology and prenatal medicine, these measurements help identify impaired placental circulation, monitor fetal growth, and guide clinical decisions when pregnancy complications affect oxygen and nutrient delivery.

Isolation of Human Umbilical Vein Endothelial Cells and Their Use in the Study of Neutrophil Transmigration Under Flow Conditions

This article first describes a procedure for isolating human endothelial cells from umbilical veins and then shows how to use these cells to examine neutrophil transmigration under flow conditions. By using a low-volume flow chamber made from a polymer with the optical characteristics of glass, live-cell fluorescent imaging of rare cell populations is also possible.

This video protocol illustrates the isolation and culture of human umbilical vein endothelial cells (HUVEC) from human umbilical cord. Once isolated these cells can be used for in vitro angiogenesis assays like the Optimized Fibrin Gel Bead Assay also demonstrated by the Hughes lab.

Coronary flow reserve (CFR) is useful for assessment of myocardial oxygen demand and evaluation of cardiovascular risk. This study establishes a step-by-step transthoracic Doppler echocardiographic (TTDE) method for longitudinal monitoring of the changes in CFR, as measured from coronary artery in mice, under the experimental pressure overload of aortic banding.

Non-invasive assessment of endothelial function in humans can be determined by the flow-mediated dilation technique. Although thousands of studies have used this technique, no study has performed this technique non-invasively in rats. The following article describes non-invasive measurement of flow-mediated dilation in the brachial and superficial femoral arteries of rats.
