Autonomic Ganglia

Autonomic ganglia are clusters of neuron cell bodies in the peripheral nervous system that relay and regulate signals between the central nervous system and involuntary organs. In sympathetic and parasympathetic pathways, preganglionic neurons release acetylcholine onto nicotinic receptors on postganglionic neurons; these neurons then transmit signals to targets such as smooth muscle, cardiac muscle, and glands, while enteric ganglia help coordinate gastrointestinal activity. Studying autonomic ganglia clarifies how the nervous system controls heart rate, blood pressure, digestion, and other homeostatic functions. Their organization and signaling are also relevant to research on autonomic disorders, pharmacology, and neural circuit development.

Dissection of Pelvic Autonomic Ganglia and Associated Nerves in Male and Female Rats

The major pelvic ganglia contain parasympathetic and sympathetic neurons that innervate pelvic organs. Here we describe a dissection method and provide schematics for identification of these ganglia and their associated nerves. These methods can be applied to experimental manipulation of these ganglia in vivo or removal post-mortem for further study.

Quantitative Autonomic Testing

Standardized, comprehensive and fully quantitative testing of autonomic functions is described. The autonomic tests consist of evaluation of all three major autonomic domains including cardiovagal, adrenergic and sudomotor. The severity and distribution of dysautonomia is quantitated using Composite Autonomic Severity Scores.

Isolation of Cells from Sensory Ganglia

This video demonstrates the isolation of cells from the sensory ganglia of mice. Ganglia are subjected to enzymatic digestion to dissolve the extracellular matrix. Single cells are then collected via filtering through a cell strainer followed by density gradient centrifugation for debris removal. Finally, washing and centrifugation are done to collect the isolated cells for future analysis.
