Auxotroph Generation

Auxotroph generation is the creation of organisms that cannot synthesize an essential metabolite and therefore require that nutrient from their growth medium. It typically involves disrupting or altering a biosynthetic gene through mutation, targeted gene replacement, or genome editing, followed by selection for cells that grow only when the missing compound is supplied. Researchers use auxotrophic strains to study metabolic pathways, control microbial growth, and introduce selectable genetic markers into engineered organisms. In biotechnology, these nutrient dependencies can improve biological containment and support the production of metabolites, recombinant proteins, and other valuable products.

Generation of Enterobacter sp. YSU Auxotrophs Using Transposon Mutagenesis

Enterobacter sp. YSU grows in glucose minimal salts medium. Auxotrophs are generated by transforming it with a transposome which randomly inserts itself into the host genome. Mutants are found by replica plating from complex medium to minimal medium. Interrupted genes are identified by gene rescue and sequencing.

Here, we present a protocol for intravital imaging of an attenuated BSL-2-approved Mycobacterium tuberculosis. strain in the murine lung using a permanent lung window. This approach enables longitudinal, single-cell-resolution analysis of early host-pathogen interactions, including bacillary localization, vascular dynamics, and macrophage uptake within the same lung microenvironment.
