Bio-rad Duoflow System

The Bio-Rad DuoFlow System is an automated liquid chromatography workstation used to separate, purify, and analyze proteins and other biomolecules. Its pumps deliver programmed buffer gradients through chromatography columns, while valves direct flow and detectors monitor signals such as ultraviolet absorbance and conductivity as components elute. Researchers can configure the system for methods including affinity, ion-exchange, and size-exclusion chromatography, adjusting flow rates and collection conditions to match the sample and column. In biology, the platform supports reproducible protein purification, method development, and fraction collection, helping investigators obtain cleaner samples for biochemical assays, structural studies, and downstream characterization.

Here we present a protocol to illustrate the isolation of rare cancer cells from a contrived sample containing whole blood and spiked in lung cancer cells. The protocol utilizes a semi-automated, label-free cell isolation platform. Enriched cells are subsequently analyzed for epidermal growth factor receptor mutations using droplet digital PCR.

This procedure describes a quick and easy workflow to introduce siRNA into difficult to transfect cell lines and follow gene expression by real-time PCR. Use of an automated cell counter, multi-well electroporation plate, and automated electrophoresis station provide quick and reliable results without the need for expensive robotic handling.
