Biotinylated Dextrin Amine

Biotinylated dextrin amine is a polysaccharide-based neuronal tracer used to map connections between regions of the nervous system. After uptake by neurons, the amine-linked polymer is transported along axons, while its biotin groups enable visualization through avidin or streptavidin conjugated to fluorescent or enzymatic labels. Researchers use this tracer to identify anterograde and, depending on experimental conditions, retrograde projections in fixed brain tissue. Labeling patterns can reveal pathway organization, terminal fields, and connectivity changes following development, injury, or experimental manipulation, supporting studies of neural circuits and the structural basis of behavior.

Transsynaptic tracing has become a powerful tool for analyzing central efferents regulating peripheral targets through multi-synaptic circuits. Here we present a protocol that exploits the transsynaptic pseudorabies virus to identify and localize a functional brain circuit, followed by classical tract tracing techniques to validate specific connections in the circuit between identified groups of neurons.
