Bisulfite Rna Sequencing

Bisulfite RNA sequencing is a method for mapping 5-methylcytosine (m5C) modifications across RNA molecules, providing insight into post-transcriptional regulation and RNA function. During the procedure, sodium bisulfite converts unmodified cytosine residues to uracil, whereas methylated cytosines resist conversion; after reverse transcription and sequencing, C-to-T differences reveal methylation sites. Applied to transcriptome-wide analyses, this approach helps identify modified transcripts, compare RNA methylation patterns between biological conditions, and investigate links between m5C, RNA stability, translation, development, and disease. Its results can also support studies of RNA-processing enzymes and broader epitranscriptomic regulation.
