Blood Velocity Measurement

Blood velocity measurement is the quantitative assessment of how rapidly blood moves through vessels, an important indicator of circulation and vascular function. In Doppler-based measurements, ultrasound interacts with moving red blood cells, and the frequency shift of reflected waves is used to calculate the component of velocity along the beam; other approaches may track labeled particles or analyze blood flow in imaging data. Measurements can be compared across vessels, physiological conditions, or disease models to characterize perfusion and hemodynamic changes. In biology and biomedical research, these data support studies of cardiovascular function, vessel development, and responses to drugs or injury.

Automated Measurement of Microcirculatory Blood Flow Velocity in Pulmonary Metastases of Rats

A method is presented to measure microcirculatory blood flow velocity in pulmonary cancer metastases of the pleural surface in rats in an automated fashion, using closed-chest pulmonary intravital microscopy. This model has potential to be used as a widespread tool to perform physiologic research on pulmonary metastases in rodents.

Micro-particle image velocimetry (μPIV) is used to visualize paired images of micro particles seeded in blood flows which are cross-correlated to give an accurate velocity profile. Shear rate, maximum velocity, velocity profile shape, and flow rate, each of which has clinical applications, can be derived from these measurements.

The focus of this manuscript is to illustrate the methods used for making precise, spatially-resolved ultrasound measurements while limiting oxidation and controlling and monitoring temperature, applied current, and the heater power being supplied.

This protocol details an assay designed to measure human neutrophil chemotaxis from one droplet of whole blood with robust reproducibility. This approach circumvents the need for neutrophil separation and requires only a few minutes of assay preparation time. The microfluidic chip enables the repeated measure of neutrophil chemotaxis over time in infants or small mammals, where sample volume is limited.
